A multicellular systems biology model predicts epidermal morphology, kinetics and Ca2+ flow

چکیده
MOTIVATION Systems biology is currently focused on integrating intracellular networks, although clinically, diseases are largely defined by their histological features. For example, no computational model can simulate today the formation of a horizontally layered epidermis. Since the epidermis is the most complex structured epithelial tissue, systems biology models could yield important insights in epithelial tissue, in which most of all human cancers arise. RESULTS We describe the algorithms of a system, capable of simulating the tissue homeostasis in human epidermis leading to a horizontally layered tissue with cells of different differentiation stages. The system predicts epidermal morphology, tissue kinetics and 2D flow of Ca2+ ions. Predicted properties of an epidermis with a healthy and a disturbed barrier are compared with the literature. The system closely mimics the respecting physiological situations. AVAILABILITY Additional information and films of the simulation are available at the website. A 3D self-organizing multicellular epidermis model of barrier formation and hydration with realistic cell morphology based on EPISIM
The epidermis and the stratum corneum (SC) as its outermost layer have evolved to protect the body from evaporative water loss to the environment. To morphologically represent the extremely flattened cells of the SC - and thereby the epidermal barrier - in a multicellular computational model, we developed a 3D biomechanical model (BM) based on ellipsoid cell shapes. We integrated the BM in the ...
متن کاملThe Potential of Menstrual Blood-Derived Stem Cells in Differentiation to Epidermal Lineage: A Preliminary Report
BACKGROUND Menstrual blood-derived stem cells (MenSCs) are a novel source of stem cells that can be easily isolated non-invasively from female volunteered donor without ethical consideration. These mesenchymal-like stem cells have high rate of proliferation and possess multi lineage differentiation potency. This study was undertaken to isolate the MenSCs and assess their potential in differenti...
متن کاملModelling Anaerobic Digestion of Cow Manure to Predict Methane Flow Rate
Anaerobic digestion (AD) of biowastes is one of the most common ways to produce methane-rich biogas, which has considerable potential to replace the fossil fuel used in multiple applications, such as vehicular transportation, internal combustion engines, cogeneration of heat and power systems and many other systems. Many companies are involved in the design and construction of anaerobic digesti...

Heavy metal pollution dispersion simulation in rivers and predicting spatial and temporal variations of pollutants can be used to determine the precise place and to schedule water withdrawal time for drinking, agriculture, aquaculture and ecosystem studies. To study the movement of heavy metal pollution through Karoon flow model, MIKE 11 was employed fpr simulation of the flow model of Karoon R...
متن کاملA Feedback Quenched Oscillator Produces Turing Patterning with One Diffuser
Efforts to engineer synthetic gene networks that spontaneously produce patterning in multicellular ensembles have focused on Turing's original model and the "activator-inhibitor" models of Meinhardt and Gierer. Systems based on this model are notoriously difficult to engineer. We present the first demonstration that Turing pattern formation can arise in a new family of oscillator-driven gene ne...
